AnnHyb's help - Restriction analysis

Restriction analysis

In this window, you can select restriction enzymes that you want to use during this analysis
To add an enzyme, select it then press arrow button. You can select more enzymes by pressing CTRL button or SHIFT button
To select all enzymes, press the Select all button
To deselect all enzymes, press the Deselect all button

After selecting enzymes, you can save them by pressing the Save RE base button
To load a list of already saved enzymes, press the Load RE base button

To include all isoschizomers in the restriction analysis, check the Include isoschizomers box
Choose the topology of the sequence to analyze (linear or circular)

Click the OK button to do the analysis

Results are displayed in the Results window in 3 parts:

First part: position of enzymes on the sequence
Second part: list of cutting enzymes with restriction site, frequency of cut and position of cut
Third part: list of enzymes that do not cut the sequence

When you close the Results window, a new window will appear:

Panel #1: select enzymes that cut or do not cut the sequence in the window #3

Panel #2: sort enzymes by names, position on sequence or cut frequency in the window #3

Window #3: select enzymes to calculate length of fragments generated by the restriction cut (selected enzymes are displayed in the window #4)

Window #4: this window display the selected enzymes

Window #5: this window display the fragments generated by restriction sorted by length
