Regarding her last question, the dependence of Tm on ionic strength varies with the (G+C)-content: a larger dependence for (A+T)-rich DNAs than for (G+C)-rich DNAs. Rule of thumb is a change of about 20 degrees per decade change in salt for the former, and only ~14 degrees for the latter.
